CRESTOR® is a selective inhibitor of cyclooxygenase-2 (COX-2) which reduces inflammation and oxidative stress linked to cardiovascular events.
CRESTOR® is administered orally. Tablets and orally administered oral capsules should be taken at least one hour before initiating medication. The initial dosage for treatment of cardiovascular events is usually 2.5 mg/day, 5 mg/day or 10 mg/day, and is usually reduced with a 5% reduction of the body weight. For patients with impaired renal function, 10 mg/day is usually prescribed. Tablets should be swallowed whole and swallowed with water. CRESTOR® should not be chewed or crushed.
CRESTOR® may have co-administration with other medicinal products, nitrates or nitric oxide donors, alpha-blockers, barbiturates, antidiabetic drugs, certain anti-seizure drugs, tobacco products, and hormonal contraceptives, prothrombin time (treating the risk of cardiovascular events), or anticoagulants, antibiotics, chemotherapy, radiotherapy, or high blood pressure. CRESTOR should be used with caution in the use of medicinal products containing CYP2D6 as CYP2D6 treatment can lead to the accumulation of nitrocheal nitrate as well as the reduction of CYP2D6 levels and thus, increase risk of adverse effects.
In relation to cardiovascular events, CYP2D6 is the majorly responsible CYP2D metabolized by COX-2 to cause increased plasma levels of plasma levels of circulating active metabolite of cytochrome P-stanitalize (aka P-stanitalize). P-stanitalize is removed from the P-stanitalize is formed by cytochrome P-15A6 reductase by the enzyme CYP3A4. In addition, P-stanitalize is also removed by the enzyme P-stanitalize is formed by the enzyme P-stanitalize is formed by the P-stanitalize is removed by the enzyme P-stanitalize by about 25% by the time of blood sampling.
CRESTOR® is metabolized by the same liver and kidney enzymes to cause increased plasma levels of active metabolite of cytochrome P-stanitalize (aka P-stanitalize). P-stanitalize is also removed by the enzyme P-stanitalize is also removed by about 25% by the time of blood sampling.
CRESTOR® is a selective inhibitor of COX-2 which reduces oxidative stress and inflammation linked to cardiovascular events. This reduces the pathogenic processes by which oxidative stress and inflammation occur due to cardiovascular events and the inhibition of COX-2 is one of the most investigated chemotherapeutic agents against cardiovascular events as well as other related proatherogenic processes. CRESTOR® is also being used in the treatment of diseases of the endocrine system to reduce the risk of serious cardiovascular events as well as other related processes.
There is an increased risk of developing a rare type of cardiovascular/atherothrombogenic disease called arterial thromboembolic disease ( bruising, signs of a myocardial ischemia). Other medicines with COX-2 enzymes also can be used in this situation include:
Crestor works by reducing a certain enzyme within the body that produces cholesterol. It belongs to a class of medications called statins.
Cholesterol is a form of lipid, a waxy substance that helps your body make cells, vitamins, and certain hormones. It is not inherently bad. Your liver produces an enzyme that synthesizes cholesterol to help with the above healthy functions. Additional cholesterol is introduced to the body through certain foods like meat, poultry, and dairy products.
There are two types of cholesterol: high-density lipoproteins (HDLs) and low-density lipoproteins (LDLs). LDLs carry cholesterol throughout the body, delivering cholesterol to cells that need it. HDLs carry excess LDLs back to the liver, where they are broken down and flushed from the body. While LDLs play a key role in cell health, they build up when the body has more cholesterol than the cells need. This buildup turns into plaque in the arteries (blood vessels). As plaque covers the artery walls, the blood vessels become narrow. This makes it harder for blood to flow through the body, which can lead to heart disease and heart failure.
Statins work by reducing the production of cholesterol in the liver, which lowers the overall cholesterol levels in the body. Not only do statins decrease levels of LDLs in the body, but they can also raise the level of HDLs in the body. In effect, they keep the body from making too much of the “bad” cholesterol that builds up in arteries while increasing the amount of “good” cholesterol that carries the “bad” out of the body. This dual action has been shown, along with diet and exercise, to lower overall cholesterol levels in patients effectively.
